چکیده

INTRODUCTION Nature has produced the most selective and efficient catalysts known, often in the form of enzymes having well defined structures to direct substrate recognition and subsequent catalysis of chemical transformations. These capabilities have inspired multiple research efforts focused on the development of supramolecular assemblies that mimic enzymes by serving as templates for chemical reactions. Pederson was among the first to demonstrate non-enzymatic reaction templation by his use of a potassium ion to direct the cyclization of benzo[18]crown-6 (Figure 1). With the development of more complex templation motifs, the definition of templation has emerged as the use of a supramolecular assembly to organize reactants in a specific spatial arrangement, favoring formation of a single product where the possibility to form more than one product exists. One motif that has recently received increased attention due to its versatility in templating chemical reactions is single stranded DNA. O

Promoting strand exchange in a DNA-templated transfer reaction.

Most DNA-templated reactions suffer from product inhibition. We explored a DNA-triggered fluorophor transfer reaction and demonstrated that comb-type polylysine-polydextran copolymers increase the turnover in template by promoting strand exchange.

Expanding the reaction scope of DNA-templated synthesis.

The translation of amplifiable information into chemical structure is a key component of nature×s approach to generating functional molecules. The ribosome accomplishes this feat by catalyzing the translation of RNA sequences into proteins. Developing general methods to translate amplifiable information carriers into synthetic molecules may enable chemists to evolve non-natural molecules in a m...

Detection of bond formations by DNA-programmed chemical reactions and PCR amplification.

A system capable of performing both DNA-templated chemical reactions and detection of bond formations is reported. Photocleavable DNA templates direct reactions. Products from bond-forming events re-ligate original templates, amplifiable by PCR, therefore distinguishing bond formation from background. This system provides a novel approach for discovering potential new chemical reactions.

Stereoselectivity in DNA-templated organic synthesis and its origins.

DNA-templated synthesis is a surprisingly general strategy for controlling chemical reactivity that enables synthetic products to be manipulated in ways previously available only to biological macromolecules. The chiral nature of the DNA template raises the possibility that DNA-templated synthesis can proceed stereoselectively. Here, we show that DNA-templated substitution reactions can exhibit...

Two enabling architectures for DNA-templated organic synthesis.

DNA templates can direct a wide variety of chemical reactions[1–24] without obvious structural requirements[23–25] by sequence specifically recruiting reactants linked to complementary oligonucleotides. The generality of DNA-templated synthesis raises the possibility of applying powerful processes such as in vitro selection and PCR (polymerase chain reaction) amplification to the discovery of s...

Translation of DNA into synthetic N-acyloxazolidines.

The translation of DNA into synthetic molecules enables their manipulation by powerful evolution-based methods previously available only to proteins and nucleic acids. The development of increasingly sophisticated DNA-templated small-molecule syntheses is crucial to broadening the scope of this approach. Here, we report the translation of DNA templates into monocyclic and bicyclic N-acyloxazoli...
